I’m using the master router to hand out DHCP assignments to manage quotas, so I have to enforce DHCP assignments. Am I correct in understanding that the checkbox simply does a double-check of DHCP assignments to prevent MAC spoofing? Otherwise MACs are still assigned IP addresses based on the table?

Are you connecting only the modem and first router via cable? Everything else is connected wirelessly?

Those are the quota statuses, not the configuration for the quota itself. This commonly happens with multiple tabs/windows open. You unfortunately can’t recover them at this point. I've had this happen a few times also, and had to rebuild the quota list. Can you give any more specifics on behavior ...
